I've got a 2000 s10 xcab, 4.3. 5 speed. The service engine soon light is constantly on due to cylinder 1 misfire. When I drove it today any gear after 1st the service engine soon light flashes. What could it be. Still only throws cylinder 1 misfire.

Start out by checking the cap and rotor (AC Delco), as well as plug wire 1 and the plug. Switch the wire and plug to another cylinder to see if it follows.
If that goes nowhere you may have a problem with that injector.

All that's already been replaced, compression tested, and noide light tested. Everything things good. I think my injector is clogged.

random only means you have more than one misfire !!! like everyone else said stick with the basics , check fire from the cap to each wire , then from the wire to he plug , then pull your plugs and make sure they are firing and not fouled ,,,you really should have a good scan test also you may have another problem causing these issues ie: bad 02 ,vacuum leak or a bad spider ,the steps above will tell you if its a spider or not
