Is anyone else having this problem on 2000 Sonoma/S10 this started yesterday while driving. This seems to be a common problem, but unsure what the cause may be. If anyone has any ideas on what may be the cause it would be greatly appreciated.

Welcome to the forum.
I'd start with new plugs and wires. Use ACDelco OEM or don't bother. These things are kinda picky that way. If that doesn't do it, do a compression test. Or do one while you have the plugs out to verify everything mechanical is ok. Getting to #3 is a lot easier if you disconnect the steering shaft. One bolt at the steering box. Then slide it up and over out of the way. Lay all the plugs out in order as you remove them and see if #5 looks different.
Next would be cap and rotor. Delco again.
